Magnesium glycinate makes you sleepy because it's doing two separate calming jobs at once. The magnesium supports GABA signalling and helps regulate your stress-hormone (HPA) axis, while the glycine it's bound to is independently calming, acting on inhibitory glycine receptors in the brain and lowering core body temperature slightly, a known trigger for sleep onset (11, 12, 13, 14). Most other magnesium forms only give you the first half of that effect.

Quick answer: It's a dual mechanism,  magnesium calms neuronal excitability and supports the stress-hormone axis, while glycine independently promotes sleep onset by lowering core body temperature and acting on its own inhibitory receptors. Together they produce a noticeably more sedating effect than magnesium citrate or oxide at an equivalent dose. Take 200–400mg elemental magnesium as glycinate, 30–60 minutes before bed.

Critical Safety Note: The sleepiness effect is generally mild and non-habit-forming, but if you feel excessively drowsy the next morning, reduce the dose or take it earlier in the evening. Check with your GP if you take sedative medication or have kidney disease.

If you've noticed that magnesium glycinate specifically seems to make you sleepy in a way other magnesium supplements haven't, you're not imagining a placebo effect, there's a genuine, two-part mechanism behind it, and understanding it properly explains a lot about why this particular form has become the go-to recommendation for sleep in clinical practice.

It's Not Just the Magnesium

Most people assume the sleepiness comes purely from the magnesium. It's true that magnesium plays a real role here: it acts as a natural antagonist at NMDA receptors (which are excitatory) and supports GABA-A receptor function (which is inhibitory), effectively turning down neuronal excitability across the central nervous system. A double-blind, placebo-controlled trial in elderly adults with primary insomnia found magnesium supplementation improved insomnia severity index scores and sleep efficiency over an 8-week period compared with placebo (8), though a subsequent systematic review notes the overall evidence base remains mixed depending on population and outcome measure (9), a fair caveat rather than a reason to dismiss the mechanism. Magnesium also has a calming effect on the HPA axis, blunting cortisol release, a topic I explore further in our piece on adrenal fatigue and cortisol (5).

But magnesium citrate, magnesium oxide and magnesium malate all deliver magnesium too, and people don't typically report the same distinctly sleepy effect from those forms at an equivalent elemental dose (15, 16) , a difference I go into in more detail when comparing magnesium forms for sleep specifically (1). That difference comes down to what magnesium is bound to, and this is where the second half of the story comes in.

Glycine Is Doing Its Own Job

In a glycinate (bisglycinate) chelate, magnesium is bound to glycine, a simple amino acid and glycine has a well-documented calming effect entirely independent of the magnesium it's attached to. It's one of the smallest amino acids in the body, and despite its simplicity, it plays a surprisingly central role in both excitatory and inhibitory neurotransmission, acting on two entirely different receptor systems depending on where in the nervous system it's released.

Glycine acts on inhibitory glycine receptors concentrated in the brainstem and, specifically, in the suprachiasmatic nucleus, the brain's master circadian clock, where NMDA receptor activation by glycine has been shown to increase blood flow and decrease nocturnal core body temperature (11). This temperature drop matters more than it might sound: the onset of sleep in humans is reliably preceded by a small but measurable decline in core body temperature, and anything that nudges that process along tends to shorten the time it takes to actually fall asleep.

Separately, oral glycine supplementation given on its own, with no magnesium at all, has been shown in a randomised, double-blinded, polysomnography-monitored trial to shorten sleep onset latency, improve subjective sleep quality, and lessen daytime sleepiness (12, 13). A related trial found glycine reduced next-day fatigue in individuals whose sleep had been experimentally restricted (14), and a study using glycine-rich collagen peptides found similar reductions in nighttime awakenings alongside lower core temperature during sleep (13). None of these studies involved magnesium whatsoever,  they isolate glycine's effect entirely, which is exactly why we can say with confidence that it's contributing something distinct from whatever the magnesium itself is doing.

So when you take magnesium glycinate, you're getting two calming mechanisms operating on two different receptor systems at the same time: magnesium's effect on NMDA/GABA-A signalling and the HPA axis, plus glycine's independent effect on inhibitory glycine receptors and core temperature regulation. That combination is why the sleepiness feels more pronounced than with other magnesium salts, and it's a genuinely useful thing to understand if you've ever wondered why switching magnesium brands or forms changed how you felt at bedtime.

Why This Matters for Choosing a Supplement

This is the exact reasoning behind why we chose magnesium glycinate, rather than a cheaper oxide or citrate, for our Sleep State formula (7). A dedicated randomised, placebo-controlled trial of magnesium bisglycinate specifically found genuine improvements in insomnia symptoms in adults reporting poor sleep (10), which is a more direct line of evidence than studies using generic "magnesium" without specifying the chelate, a distinction that matters more than most supplement labelling makes clear, since so many trials in this space use whichever magnesium salt happened to be convenient rather than the one most relevant to sleep specifically.

A Practical Note on Dose and Timing

Most people notice the calming effect within 30–60 minutes, which is why it's best taken in that window before bed rather than with dinner several hours earlier. The typical effective range is 200–400mg of elemental magnesium, worth checking the label carefully, since "magnesium glycinate 800mg" often refers to the total weight of the compound rather than the elemental magnesium it actually contains, which is usually a smaller fraction of that figure.

If you wake up still groggy, that's usually a sign the dose was slightly too high for you or taken too late,  reducing it, or shifting it earlier in the evening, tends to resolve this without losing the sleep benefit. It's also worth noting that the glycine component means this particular form may feel more noticeably sedating at a lower elemental dose than you'd expect from magnesium alone, so it's reasonable to start conservatively and adjust upward only if needed.

When It's Not Just Magnesium You Need

Magnesium glycinate is a genuinely well-evidenced tool, but it's one part of a bedtime routine, not a substitute for one. It's also worth remembering that magnesium and glycine both work through nervous-system calming rather than sedation in the pharmaceutical sense, they lower the threshold for sleep to occur naturally, rather than forcing unconsciousness the way a hypnotic drug does, which is part of why the effect tends to feel gentler and why there's no evidence of the tolerance or dependence associated with some sleep medications.

The Big Picture

Magnesium glycinate makes you sleepy because it's quite literally doing two jobs at once,  magnesium calming the nervous system and regulating stress hormones, glycine independently lowering core temperature and supporting inhibitory signalling in the brain. That dual mechanism is the practical reason it outperforms other magnesium forms specifically at bedtime, and why it's worth choosing deliberately rather than reaching for whichever magnesium happens to be cheapest on the shelf.
