It's Not Just the Magnesium

Most people assume the sleepiness comes purely from the magnesium. It's true that magnesium plays a real role here: it acts as a natural antagonist at NMDA receptors (which are excitatory) and supports GABA-A receptor function (which is inhibitory), effectively turning down neuronal excitability across the central nervous system. A double-blind, placebo-controlled trial in elderly adults with primary insomnia found magnesium supplementation improved insomnia severity index scores and sleep efficiency over an 8-week period compared with placebo (8), though a subsequent systematic review notes the overall evidence base remains mixed depending on population and outcome measure (9), a fair caveat rather than a reason to dismiss the mechanism. But magnesium citrate, magnesium oxide and magnesium malate all deliver magnesium too, and people don't typically report the same distinctly sleepy effect from those forms at an equivalent elemental dose (15, 16), a difference I go into in more detail when comparing magnesium forms for sleep specifically (1). That difference comes down to what magnesium is bound to, and this is where the second half of the story comes in.

Glycine Is The Primary Sleep Agent

In a glycinate (bisglycinate) chelate, magnesium is bound to glycine, a simple amino acid and glycine has a well-documented calming effect entirely independent of the magnesium it's attached to. It's one of the smallest amino acids in the body, and despite its simplicity, it plays a surprisingly central role in both excitatory and inhibitory neurotransmission, acting on two entirely different receptor systems depending on where in the nervous system it's released.

Glycine acts on inhibitory glycine receptors concentrated in the brainstem and, specifically, in the suprachiasmatic nucleus, the brain's master circadian clock, where NMDA receptor activation by glycine has been shown to increase blood flow and decrease nocturnal core body temperature (11). This temperature drop matters more than it might sound: the onset of sleep in humans is reliably preceded by a small but measurable decline in core body temperature, and anything that nudges that process along tends to shorten the time it takes to actually fall asleep.

Separately, oral glycine supplementation given on its own, with no magnesium at all, has been shown in a randomised, double-blinded, polysomnography-monitored trial to shorten sleep onset latency, improve subjective sleep quality, and lessen daytime sleepiness (12, 13). A related trial found glycine reduced next-day fatigue in individuals whose sleep had been experimentally restricted (14), and a study using glycine-rich collagen peptides found similar reductions in nighttime awakenings alongside lower core temperature during sleep (13). None of these studies involved magnesium whatsoever,  they isolate glycine's effect entirely, which is exactly why we can say with confidence that it's contributing something distinct from whatever the magnesium itself is doing.

So when you take magnesium glycinate, you're getting two calming mechanisms operating on two different receptor systems at the same time: magnesium's effect on NMDA/GABA-A signalling and the HPA axis, plus glycine's independent effect on inhibitory glycine receptors and core temperature regulation. That combination is why the sleepiness feels more pronounced than with other magnesium salts, and it's a genuinely useful thing to understand if you've ever wondered why switching magnesium brands or forms changed how you felt at bedtime.

Why Glycine Lowers Your Core Body Temperature

The temperature mechanism is worth explaining properly, because it's the part most articles gloss over and it explains a lot about how the effect actually feels.

Human sleep onset is preceded by a small drop in core body temperature — usually in the region of 0.3 to 0.4°C. This isn't incidental to falling asleep; it's part of the signalling that initiates it. Your body achieves that drop not by generating less heat but by shedding more of it, widening the blood vessels in your hands and feet so warmth radiates away from the core. It's why cold extremities at bedtime are associated with difficulty falling asleep, and why a warm bath an hour or two before bed helps: the subsequent rebound cooling amplifies the same signal.

Glycine appears to work directly on this system. Research in animal models has shown that glycine acts at NMDA receptors in the suprachiasmatic nucleus, the hypothalamic structure that governs circadian rhythm, increasing blood flow to the periphery and producing a measurable fall in core temperature (11). A human study using glycine-rich collagen peptides found lower core temperature during sleep alongside fewer nighttime awakenings (13).

So the glycine in magnesium glycinate isn't simply calming in a general sense. It's nudging a specific physiological switch that your body would otherwise be flipping on its own schedule. That's a meaningfully different thing from muscle relaxation, and it's the clearest reason why this particular chelate behaves differently at bedtime from other magnesium salts.

Does Magnesium Glycinate Make You Sleepy Immediately?

Not immediately, but sooner than most supplements. Most people notice a settling effect within 30 to 60 minutes, and there's a reason that window is fairly consistent.

Glycine is one of the smallest amino acids in the body and it's absorbed quickly, with plasma concentrations typically peaking within the first hour of an oral dose. The magnesium component moves more slowly, since mineral absorption across the gut wall is a more gradual process, but the glycine reaching circulation early is doing much of the initial work — which is part of why the glycinate form tends to feel faster than magnesium citrate or oxide.

What you won't feel is a sudden onset. Magnesium glycinate doesn't sedate in the pharmaceutical sense; it lowers the threshold at which sleep can happen naturally. The sensation people describe is usually a gradual softening, muscles letting go, the mental chatter quietening rather than the abrupt heaviness of a hypnotic. If you're expecting the latter, you may conclude it isn't working when it is.

The practical consequence is that timing matters more than it would with a fast-acting sedative. Taken with dinner at seven for a bedtime of eleven, the most noticeable part of the effect has often passed before you get into bed.

Why This Matters for Choosing a Supplement

A dedicated randomised, placebo-controlled trial of magnesium bisglycinate specifically found genuine improvements in insomnia symptoms in adults reporting poor sleep (10), which is a more direct line of evidence than studies using generic "magnesium" without specifying the chelate — a distinction that matters more than most supplement labelling makes clear, since so many trials in this space use whichever magnesium salt happened to be convenient rather than the one most relevant to sleep specifically.

Best Time to Take Magnesium Glycinate: Morning or Night?

For sleep, take it in the evening — 30 to 60 minutes before bed, which puts peak glycine absorption roughly where you want it.

Whether it needs to be evening at all depends on what you're taking it for. Magnesium status in the body is cumulative rather than acute; you're topping up a mineral pool, not dosing a drug. If your reason for supplementing is general magnesium repletion — muscle function, energy metabolism, nervous system support — the time of day makes very little difference, and morning is perfectly reasonable.

The evening timing matters specifically for the sleep effect, because that's driven by glycine, which clears from circulation within a few hours. That's a same-night mechanism, not a cumulative one.

Three situations where I'd suggest morning instead:

If it leaves you groggy the next day. Shifting the dose earlier usually resolves this without losing the benefit.

If it makes you more alert rather than less. This is uncommon but real, and it's covered in the section on paradoxical responses below.

If you're splitting a larger dose. Some people tolerate magnesium better in two smaller amounts than one large one, in which case morning and evening works well.

There's no evidence that taking magnesium in the morning disrupts sleep architecture that night, and no reason to worry if your routine suits it better.

Can You Take Magnesium Glycinate on an Empty Stomach?

Yes, and it's one of the practical advantages of this form.

Magnesium oxide and magnesium citrate are both known for causing gastrointestinal upset: loose stools, cramping, nausea, particularly when taken without food. Magnesium citrate is used as a laxative at higher doses for exactly this reason. In a glycinate chelate, the magnesium is bound to two glycine molecules, and the resulting compound appears to be absorbed partly through amino acid transport pathways rather than relying entirely on the mineral absorption routes that draw water into the bowel. In practice that means far fewer people report digestive symptoms, empty stomach or not.

Taking it with food is still fine. There's a modest theoretical reduction in absorption when minerals compete with other dietary components — calcium and zinc in particular — but the difference isn't large enough to matter for most people, and consistency is worth more than optimization.

The one thing I'd avoid is taking it alongside a high-dose calcium or iron supplement, since those do compete meaningfully for the same transporters. Separate them by a couple of hours.

If Magnesium Glycinate Keeps You Awake

Paradoxically, some people report feeling more alert after taking magnesium glycinate before bed, and the human evidence explaining this response is limited. The likeliest mechanism sits with the glycine rather than the magnesium. Glycine acts as an inhibitory neurotransmitter at glycine receptors in the brainstem and spinal cord, which is part of why it generally supports sleep. It also acts as a co-agonist at NMDA receptors, where it is required alongside glutamate for excitatory signalling. In a nervous system already running hot, nudging that second pathway may be enough to tip the balance towards arousal rather than rest.

Context matters as much as chemistry. It is my clinical experience that people seeking magnesium glycinate for sleep frequently present with a dysregulated HPA axis and cortisol that stays higher than it should into the evening. A supplement taken at 10pm does not create that pattern, but it is easily blamed for it. Caffeine has a half-life of roughly five to six hours, so a mid-afternoon coffee is still circulating at bedtime. Evening exercise elevates core temperature and catecholamines for some hours afterwards. Most of us also pass through a natural wake-maintenance zone in the one to three hours before habitual bedtime, when circadian drive for alertness peaks just ahead of melatonin release. Any of these can produce a burst of energy that simply coincides with a new supplement.

If this sounds familiar, shift your magnesium to the morning and start at a smaller dose while your metabolic pathways adjust. Bring your caffeine cut-off forward to midday, and move vigorous training earlier where you can. If alertness persists, a different form such as magnesium malate or taurate may suit you better, and it is worth investigating your cortisol rhythm properly.

Will Magnesium Glycinate Make You Drowsy the Next Day?

Usually not and the evidence here runs in an interesting direction.

Glycine has been studied specifically for next-day effects, and the findings are the opposite of what you might expect. A randomised trial found that glycine taken before bed reduced daytime sleepiness and improved subjective feelings of fatigue the following day (12). A separate trial in people whose sleep had been experimentally restricted found glycine lessened next-day fatigue rather than adding to it (14). If glycine were simply a sedative with a hangover, you'd see the reverse.

So when people do report morning grogginess after magnesium glycinate, it's worth being precise about the likely cause, because it usually isn't residual sedation. Glycine clears from circulation within a few hours and isn't still working on you at eight the next morning.

The more common explanations:

The dose was higher than you needed. This is the most frequent cause, and the easiest to test: reduce it and see.

You took it too late. A dose at midnight puts the calming window into the early hours rather than at sleep onset.

You slept more deeply than usual and woke mid-cycle. Waking from deep sleep produces sleep inertia, that thick-headed feeling that takes 20 to 30 minutes to clear. It's a sign the supplement worked, not that it's sedating you. It typically settles as your body adjusts over a week or two.

Something else is going on. Persistent daytime tiredness that doesn't resolve when you adjust dose and timing isn't a supplement problem. Low ferritin, thyroid dysfunction, sleep apnoea and vitamin D deficiency all present this way and all warrant a conversation with your GP rather than a change of magnesium.

If you want the sleep benefit without the morning heaviness, halve the dose for a week before changing anything else. That resolves it for most people.

Why Do I Feel So Tired After Taking Magnesium Glycinate?

If the tiredness arrives in the evening, that's the mechanism working as intended, and the answer is simply to take it closer to when you actually want to sleep.

If it arrives during the day, the question is what changed. Magnesium doesn't accumulate to sedating levels in a healthy person, the kidneys regulate it closely, excreting what isn't needed. A tired feeling that persists through daylight hours is rarely the magnesium itself.

Three possibilities worth separating:

You were magnesium-deficient and your body is recalibrating. Correcting a genuine deficiency can unmask how tired you already were, particularly if adrenaline had been compensating. This usually settles within two to three weeks.

You took it too close to waking. A dose at 6am with the intention of "getting it out of the way" puts the glycine window squarely in your morning.

The sleep itself has changed. Sleeping more deeply for the first time in a while can leave you feeling heavier rather than fresher for several days before it evens out.

What I'd take seriously is tiredness that arrives with muscle weakness, unusual shortness of breath, or a slowed heart rate. That combination is not a normal response to a supplement and warrants medical attention, particularly if you have any degree of kidney impairment, since impaired kidneys don't clear excess magnesium efficiently. This is rare in people with normal renal function taking sensible doses, but it's the one scenario where the answer isn't "adjust your timing".

When It's Not Just Magnesium You Need

Magnesium glycinate is a genuinely well-evidenced tool, but it's one part of a bedtime routine, not a substitute for one. It's also worth remembering that magnesium and glycine both work through nervous-system calming rather than sedation in the pharmaceutical sense, they lower the threshold for sleep to occur naturally, rather than forcing unconsciousness the way a hypnotic drug does, which is part of why the effect tends to feel gentler and why there's no evidence of the tolerance or dependence associated with some sleep medications.

That dual mechanism is the practical reason it outperforms other magnesium forms specifically at bedtime, and why it's worth choosing deliberately rather than reaching for whichever magnesium happens to be cheapest on the shelf.

Long term. Healthy kidneys excrete what you don't need, so magnesium doesn't accumulate at sensible doses. It's a nutrient rather than a sedative, so there's no tolerance or dependence mechanism. Most likely you're correcting a dietary shortfall, which means the benefit fades if you stop and your diet hasn't changed.

Where it goes wrong. The upper limit for supplemental magnesium is 350 mg daily, or 400 mg under UK guidance. Above that, expect diarrhoea and cramping. Reduced kidney function is the real red flag, since magnesium can then build up dangerously. It also interferes with bisphosphonates, some antibiotics, diuretics and long-term reflux medication.

Check your label. Magnesium glycinate is only about 14% elemental magnesium by weight, so a "1,000 mg" capsule delivers roughly 140 mg. Aim for 200–250 mg elemental, 30–60 minutes before bed.

You swallow a lot of it. Pure glycinate is only about 14% elemental magnesium by weight, so a 250 mg elemental dose needs roughly 1,775 mg of compound: two to four large capsules nightly. Magnesium oxide is around 60% elemental, so one small tablet does the same job. Glycinate also costs more per milligram of actual magnesium delivered.

The labels mislead. "1,000 mg magnesium glycinate" describes the whole molecule; you're getting about 140 mg of magnesium. Some brands also buffer the product with cheap magnesium oxide to inflate the front-of-bottle number, which dilutes the gentleness you paid for. Check the ingredients panel for oxide.

The benefit is modest. The largest trial found insomnia scores improved by 3.9 points against 2.3 on placebo, an effect size described as small. A meta-analysis found people fell asleep around 17 minutes faster, on evidence rated low to very low quality. The NIH's own list of better-absorbed magnesium forms doesn't include glycinate.

It can still upset your stomach. Gentler than oxide doesn't mean exempt. Diarrhoea and cramping scale with dose.

Reaching 250 mg elemental takes around 1,775 mg of compound, which is two to four large capsules. Read the supplement facts panel, not the front of the bottle.

Where 250 mg comes from. It's the dose used in the only randomised trial of magnesium bisglycinate for sleep, published in 2025. Insomnia scores improved by 3.9 points against 2.3 on placebo over four weeks. The effect was small. No trial has compared doses for sleep, so there's no evidence more works better.

How to start. Take 100–150 mg elemental for the first week to check your gut tolerates it, then move to 200–250 mg. Splitting the dose or taking it with food helps if you're sensitive.

Subtract your diet. Daily requirements cover food and supplements together: 300 mg for men and 270 mg for women in the UK, 400–420 mg and 310–320 mg in the US. If you eat plenty of greens, nuts, seeds and whole grains, you need less from a capsule. The 2025 trial found the biggest responders were those eating the least magnesium.

Stay under the ceiling. 350 mg a day from supplements in the US, 400 mg under NHS guidance. Food doesn't count, but antacids, laxatives and multivitamins do.

Review after a month. Most of the trial's improvement appeared within two weeks. If six weeks changes nothing, the dose isn't the problem. Don't supplement at all without medical advice if you have kidney problems or take bisphosphonates, antibiotics or diuretics.

What to avoid when taking magnesium glycinate?

Mostly it's a matter of timing and double-counting rather than hard prohibitions.

Space it from certain medicines. Magnesium binds to bisphosphonates and antibiotics in the gut, and to levothyroxine, which needs four hours' separation. Taking magnesium at bedtime usually solves this, since these are morning medicines. Diuretics and long-term reflux medication affect your magnesium levels directly, so ask a prescriber rather than just adjusting timing. Very high-dose zinc also interferes with absorption.

Don't double-count. The ceiling is 350 mg a day from supplements, or 400 mg under NHS guidance. Magnesium hides in antacids, laxatives, multivitamins, electrolyte powders and combination sleep blends. Add them all up first. Food doesn't count.

Avoid buffered products. Pure glycinate is about 14% elemental magnesium, so a normal capsule claiming 200 mg elemental has been blended with cheap magnesium oxide. Check the ingredients panel. Avoid labels stating only compound weight, and blends that don't disclose the amount.

Avoid a few habits. Don't pair it with a nightcap. Alcohol fragments sleep, and heavy drinking is a risk factor for low magnesium. Don't take it on an empty stomach if you're sensitive. Don't raise the dose when it isn't working, since no trial has compared doses. Don't judge it before four weeks, and don't stack several sleep aids at once.

Avoid it entirely if your kidneys are impaired. They can't clear magnesium, and advanced kidney disease risks hypermagnesaemia. Children need medical advice; their upper limits are far lower.

Above all, don't let it replace a diagnosis. Persistent poor sleep may be apnoea, restless legs or chronic insomnia, for which CBT-I outperforms any supplement. And if you take regular medication, ask a pharmacist to check your whole list. It takes two minutes.
